In the development of the pharmaceutical industry, pharmaceutical excipients play a vital role. Microcrystalline cellulose (MCC), depend on its unique physicochemical properties, has become an indispensable excipient in the pharmaceutical field, playing a key role in the production of various drug formulations.
I. The Characteristics of Microcrystalline Cellulose and Its Relevance to Pharmaceuticals
Microcrystalline cellulose is a highly crystalline product derived from natural cellulose through specific processing, primarily composed of D-glucose units linked by β-1,4-glycosidic bonds. It appears as a white or off-white powder, odorless and tasteless, and is insoluble in common organic solvents. MCC has excellent flowability and compressibility, which enable it to effectively improve the processing properties of materials in the production of solid formulations such as tablets and capsules. Meanwhile, its low hygroscopicity helps maintain the stability of drug formulations, preventing drug deterioration caused by moisture absorption. Additionally, the abundant hydroxyl groups on its surface endow it with certain chemical reactivity, and its application scope in the pharmaceutical field can be further expanded through chemical modification.
II . The Core Role of Microcrystalline Cellulose in Pharmaceutical Excipients
(1) High-Quality Bulking Agent
MCC is widely used as a bulking agent in the production of tablets and capsules. For formulations with low active ingredient content, MCC can increase the volume and weight of tablets or capsule contents to meet appropriate specification standards. Take a certain vitamin tablet as an example: since the content of the main drug (vitamin) is low, adding MCC not only fills the tablet volume but also ensures the tablet's hardness and uniform appearance, facilitating patient administration and large-scale production. Moreover, the good flowability of MCC ensures uniform distribution of materials during mixing and tableting, avoiding issues of content non-uniformity.
(2) Efficient Disintegrant
The rapid disintegration of drugs in the human gastrointestinal tract is a prerequisite for effective absorption. As a disintegrant, MCC can significantly shorten the disintegration time of tablets. When a tablet enters the gastrointestinal tract, MCC rapidly absorbs water and swells, disrupting the tablet's structure and promoting rapid drug release. In some compound formulations, the mixing of multiple drug ingredients may affect the tablet's disintegration performance, and the addition of MCC can effectively improve this situation. For instance, in a compound tablet for cold treatment containing multiple active ingredients, MCC as a disintegrant ensures rapid disintegration in the gastrointestinal tract, accelerates drug absorption, and thereby enhances the therapeutic effect.

In wet granulation processes, MCC can be used as a binder. It can tightly bind with drug powders to form granules with certain strength and particle size, improving the compressibility and formability of materials. The binding effect of MCC is particularly important for drugs that are difficult to form. For example, some natural extract-based drugs have low inherent viscosity, prone to loose granules and poor formability during granulation. Adding MCC can effectively solve these problems, improving granule quality and stability, and laying a good foundation for subsequent tableting.
